Nonstop mRNA degradation is thought to be triggered when a translating ribosome reaches the very 3' end of an mRNA. The ribosome is recognized by the C-terminal domain of Ski7p, possibly because the ribosomal A-site does not contain a codon. Recognition of the ribosome leads to 3' to 5' degradation by a large ribonuclease complex(the exosome).
